Immunology
(a) Antibody Research: Preparation of polyclonal and Monoclonal antibodies, purification and detection by immunofluorescence, immunodiffusion, radioimmunoassay, ELISA, Luminex, immunoblots, immunoprecipitation, and FACS. Lymphocyte characterization: Isolation, culture, co--‐culture, cloning, proliferation and cytotoxicity assays, ELISPOT, apoptosis, phenotype analyses by FACS and adoptive transfer experiments. (b) Animal models: Autoimmune, graft vs. host disease, transplantation, and antibody--‐mediated neurological disease in SCID, nude mice and rat models. (c) Signal transduction: Lymphocyte & Epithelial cells – GPCR, BCR and ROS. Ras/cRaf/B--‐Raf/MAPK NFkB, NFAT, Caspase/FAS, Bcl--‐2, Bcl--‐xL, Bcl--‐6, CD40/CD40L, TLR, etc. (d) Mitochondria, endoplasmic reticulum, golgi, and vesicular trafficking (endosome--‐lysosome) and sorting.
Biochemistry
Protein chemistry): Centrifugations, affinity purification, gel purification, 1-D, 2‐D gel‐electrophoresis, Western blot, EMSA, FPLC, HPLC, digestion, microsequencing, mass spectrometry, etc.
Neuroimmunology & Endocrinology
Animal models, perfusion, neuronal cell culture, immunohistochemistry (frozen sections, fixed tissue section, free floating, antigen retrieval), Immunofluorescent, protein purification from brain, cell--‐based assay, staining of astrocyte, microglia, oligodendrocytes and neurons, thyroid cell signaling, animal models of thyroid autoimmunity. TSH receptor antibody‐induced Signal transduction with kinase assays, proteomics by antibody array, and System Biology.
