Sendai viral reprogramming techniques are used to reprogram patient fibroblasts to induced pluripotent stem cells. We are continuously testing and adapting state-of-the-art neural differentiation protocols to improve the yield and purity of glutamatergic, GABAergic, and dopaminergic neurons generated from our hiPSCs. Molecular profiles of control and SZ neural progenitor cells and neurons are compared by RNAseq and microarrays. Cellular phenotypes of NPCs are identified by assays for cellular replication, death, oxidative stress, and migration. SZ neurons are phenotyping by assays for dendritic arborization, synaptic density, rabies virus transneuronal tracing, calcium imaging, and electrophysiology.
