Equipment and Rates

The Flow Cytometry CoRE is comprised of multiple machines from established manufacturers to provide the best range of cytometric capabilities and applications. To facilitate versatility, all BD analyzers and cell sorters use a detector configuration that allows for template portability, as well as preventing figures and data from being mislabeled in publication. The most common fluorochromes in each detector are listed in a table that can be used to choose the appropriate instrument for your samples.

Rates

All instrument usage rates are indicated at the point of reservation within the iLab.

Cancellation Charges

All scheduled sessions in the iLab system are subject to a 24 hour advance cancellation period. Cancellation within 24 hours of the scheduled start of a session will result in a charge equal to the entire scheduled session time. This cancellation charge will be pro-rated based upon assumption of the cancelled time by another user.

Data Workstations - No charge

The CoRE maintains PC workstations for use free-of-charge to analyze data generated within the CoRE. These workstations have up-to-date versions of FCSExpress, IDEAS, DiVa, Spectroflo, and access to cloud services such as Fluorofinder and Cytobank, as well as Microsoft Office. Other data analysis packages may be available as beta test versions predicated on Mount Sinai’s reputation for high volume flow cytometry and broad subject matter.

Five workstations are placed in IMI13-05 and two in CSM S6-308 for use on a 24/7 schedule.

Software

The CoRE provides a site license for FCS Express Version 7 Flow Cytometry analysis software. You may request a license and access additional resources to help you get started with FCS Express through the iLabs system in the Software Licenses section at the bottom of the Request Services tab.

The team at De Novo Software is committed to your success. Contact support@denovosoftware.com for immediate assistance or register for free Q&A sessions.

Below are the various types of analyzers we employ.

Canto II

Manufactured by BD, this instrument features three lasers and eight colors, as well as semi-automated fluidic operation.

LSRFortessa X-20

Manufactured by BD, this instrument features five lasers and 18 colors for maximum complexity of experimental design. This instrument has a high throughput HTS option available to read samples from 96 and 384 well microtitre plates.

Attune NxT

Manufactured by ThermoFisher, this instrument features four lasers and 14 colors for maximum complexity of experimental design.

Aurora

Manufactured by Cytek Biosciences, this instrument features five lasers and spectral detection technology to facilitate more than 60 discrete multiplexed probes per cell.

ImageStream

Manufactured by Luminex/Amnis, this instrument features six lasers and twelve spectral channels plus bright and darkfield for high speed image capture of individual cells in suspension. The number of discrete parameters per cell can be greater than fifty based on image features.

Our CoRE works with eight different cell sorters as detailed below.

CS

Manufactured by CytekBiosciences, this instrument features five lasers and full spectrum detection technology to provide up to 60 colors inside a biosafety cabinet for sorting human or potentially infectious material. This instrument is preferentially scheduled for self-operation.

SRT

Manufactured by Beckman Coulter Life Sciences, this instrument features five lasers and 15 colors inside a biosafety cabinet for sorting human or potentially infectious material. This instrument is preferentially scheduled for self-operation.

IMI5L

Manufactured by BD, this instrument features four lasers and 12 colors inside a biosafety cabinet for sorting human or potentially infectious material.

CSM5L

Manufactured by BD, this instrument features five lasers and 16 colors inside a biosafety cabinet for sorting human or potentially infectious material. This instrument is preferentially scheduled for self-operation.

Syd

Manufactured by BD, this instrument features four lasers and 16 colors inside a biosafety cabinet for sorting human or potentially infectious material. This instrument is preferentially scheduled for self-operation.

Tyto181 and Tyto183

Manufactured by Miltenyi, these identical instruments feature cartridge based microfluidic sorting for true sterility. The optics are configured with three lasers and 8 fluorescence detectors as well as three backscatter detectors. These instruments are preferentially scheduled for infectious material sorting.

Wolf

Manufactured by Nanocellect, this instrument features 3 colors and microfluidic containment for inherently biosafe sorting. Wolf is currently installed within the Department of Microbiology BSL3 laboratory and requires additional training for facility access.
